While both are definitely antagonists, I always viewed Fontaine as the main, simply because the game isn't over until he is defeated. The game switches the antagonist with a twist, revealing an even bigger antagonist who has been pulling the strings (and Jack's strings) the whole time. Also, he has the boss battle while Ryan dies just like that for the twist of revealing everything was a lie
That, and when I google Bioshock 1 main antagonist Fontaine is the result

They didn't all happen in the BioShock poll though? Andrew Ryan, the main antagonist, wasn't defaced with "WHORE" on his poster, it was Brigid Tenenbaum, and twice. Once you find the wall in her old home is defaced with it scratched into the wall, the second time was in Burial at Sea where her poster is the only one to have the face not recognisable with "WHORE" written on it.
